A friend of mine claims that using HPTuners and modifying the DS tune is possible. If a car has the DS tune installed and someone attempts to modify the tune with HPTuners or some other tuning software, would it work? How would it affect using the Predator again?

Not a good idea.
It is possible, but if you flash with HP over the Predator tune, the Predator will not recognize it and will lock you out.
