Mustang V6 - 3.73 gears

My 2007 Mustang V6 has an Airaid Cold Air Intake, Magnaflow Cat Back Dual pipes and a Diablosport High Performance (Firm shift) tune. If I swap out the gears with 3.73's, will that effect my tune?

It won't effect your tune just your speedo reading correctly. But lucky for you the predator will allow you to make the gear swap change so your speedo will read correctly. One of the benefits of owning a predator.

Will the speedo typically read faster or slower?

When you install larger gears than stock, the speedo will read that you are going faster than you actually are.
