I have a 2016 Silverado, 5.3 EcoTec. I installed CAI, Shorty JBA Headers and Magnaflow exhaust Cat' back. I'm running the Trinity 91 at the moment. Would this be sufficient or... Should I get a custom tune?

you dont NEED a custom tune...but it may help at this point
