I bought this unit for my 2018 Silverado and so far, so good. I’m wondering if I can use it on my 05 Chrysler 300C as well?

yes, add a dodge license for $165 and wake up that 300C!

Will I need to remove my truck programming from the device first, or will I be able to have access to both my truck and my 300C with the unit?

with an additional license you can use the same device to tune both vehicles (or more)
