I just want to make sure my predator u7158 will support a #6

I have a predator u7158 with a custom tune for a #4 lower pulley (approx 12lbs of boost.) Im thinking of swapping it out w/ a #6 (approx 14lbs.) Will that tune support the added 2 lbs or do I need to have a dyno custom tune done?

I have a 03 Lightning. Mods are: BBK twin 65mm TB, Bassani catback, K&N air filter, 170 stat, boost bypass kit and NGK TR6`s.

Do you mean you have a custom tune from a CMR dealer, or you are using the pulley tune from the Predator?
Either way whether or not it will handle more boost depends on what the A/F ratio is right now, and what it will be after the swap.
Best bet is to plan on getting a new tune.
