i want to install magnacharger on my 08 denali and according to the magnachager Installation Instructions i have to send my ECM and TCM for for calibration do i have to that or make a custom tune using predator

yes, you will need to do that...
I'm sure you can get a custom tune from the diablo guys that will get you close and driveable, but you'd still need to take it to a dyno/shop and get it tuned correctly for that application.
